ステロイドホルモンレセプターのヌクレオソーム結合部位を構成する蛋白質の構造解析

研究概要	グルココルチコイドレセプター(GR)情報伝達パスウェー上で必須の因子と考えられるGR核結合促進因子(ASTP)の作用機構は、核内でGRと協調してヌクレオソーム構造を変化させている可能性が考えている。本研究は、このASTPの一次構造を解析する事により、ASTPとGR、主要なヌクレオソーム構成成分であるヒストン及び特異的DNA配列(ホルモンレスポンスエレメント:HRE)の相互作用におけるASTPの役割を解明しようとしている。ASTPを電気泳動的に単一にまで精製し、リジンエンドペプチダーゼ、V8プロテアーゼで限定分解し電気泳動で分離、PVDF膜に転写して得られた6つのペプチドのN末端のアミノ酸配列をそれぞれエドマン法にて決定した。さらに精製ASTPを用いて特異的な抗ASTP抗体の作製に成功した。次に、この抗ASTP抗体を用いて、ラット肝cDNAライブラリーからASTP蛋白発現ベクターのスクリーニングを行ない、3個のクローンを得た。これらのクローンをPCR法で増幅し、得られたインサートDNAの全塩基配列を蛍光ターミネーター・シークエンス法で決定した。その結果、ASTPの全cDNA配列を解析出来た。ASTPのcDNAの全配列は2989bpで、1572bpから成るOpen Reading Frameを含み、524個のアミノ酸から成る蛋白質をコードしていた。cDNA配列より予想されるアミノ酸配列は精製ASTPの限定分解により得られた6種類のアミノ酸配列を全て含んでいた。また、その配列にはSV40のT抗原やステロイドホルモン受容体に認められる核移行シグナルに類似した配列が認められた。
