老化促進マウスを用いた造血系老化の研究

研究概要	我々は、老化促進モデルマウス(SAM-P)には、加齢に伴って末梢血球数、特に白血球数の有意な減少が見られることを、既に報告している。昨年度は、老若マウス間で骨髄相互移植を行った実験から、これが加齢マウスでみられる末梢血球の減少が造血支持環境の、細胞分化誘導機能の低下によるものであることを示めした。本年度は、この加齢に伴って低下する造血支持機能の要因を明らかにするために、老若マウス骨髄細胞からのGM-CSFの産生を調べた。幼若マウスに較べて加齢マウスでは、GM-CSFの産生量が明らかに減少していたが、LA-3刺激に反応して起る、このGM-CSFの産生誘導/亢進には差が認められなかった。しかし、これらの刺激を受けた時のGM-CSFの総産生量は幼若マウスの産生量より少ないことが判明した。なお、細菌内毒素(LPS)刺激に対する反応には、老若マウス間で差は認められなかった。これは、IL-3と違って、非造血制御因子による強い外来刺激に対する反応には、僅かな機能低下の影響がみられないためと思われる。また、IL-6の産生に関しては、老若マウスの間に有意な差はみられなかった。以上、本年度の研究によって、加齢マウスにおける末梢白血球数の減少が、造血組織の白血球誘導因子産生能の低下のために、成熟白血球までの分化が低減するという、造血支持機能の老化を主たる原因とすることが明らかになった。
