加齢に伴うマクロファージ機能の変化

研究概要

マクロファージは、感染防御や創傷治癒などの生体の恒常性の維持に重要な働きをしている。従って、加齢に伴うマクロファージ機能の変化は少なからず、成人病や老年病の発生に関与している。今回、我々はマクロファージの加齢変化は臓器の環境に依存していることを示唆する以下の結果を得た。
1.マクロファージの腫瘍壊死因子(TNF)産生能の加齢変化は、臓器によって異なる。肺胞マクロファージのTNF産生能は、加齢と共に減少するが、腹腔では増加し、骨髄では、変化しなかった。
2.単球・顆粒球幹細胞(GM-CFU)は、骨髄では増えていたが、脾臓では変化が無かった。
3.単球・マクロファージ幹細胞(M-CFU)は、骨髄では増えていたが、肺では逆に加齢と共に減少した。
以上の結果から、マクロファージおよびその幹細胞の加齢変化は、マクロファージおよびその幹細胞の加齢変化は、マクロファージの存在する臓器に依存していることが明らかとなった。
