研究概要

がん遺伝子、がん抑制遺伝子の研究が進み、がんを遺伝子ならびにその産物の機能に基づいて理解することが可能になった。それと共に、細胞増殖のためのシグナル伝達や細胞周期制御の機構の解明が進んだ。さらにこれらの知見を基にがんの新しい診断法や治療法が開発されようとしている。このような状況下、先端的がん研究の情報収集ならびに交換のために本年度は19名の班員を米国に派遣した。がん発症と進展に関するより深い理解を得るために、がん遺伝子・がん抑制遺伝子の機能解析を中心とする、シグナル伝達制御ならびにDNA合成の分子機構を含む細胞周期制御に関するテーマで9名の研究者を、また転移の仕組み等のがんの悪性化の機構解析に2名、そしてがん治療の観点から8名の研究者がこの派遣プロジェクトに組織された。特にがん遺伝子ならびにがん抑制遺伝子の機能や遺伝子導入法に基づくがん治療の分野は発展途上であり、重点的に情報収集を行う必要がある。今年度はがん遺伝子産物等のシグナル伝達で重要な役割を果たす蛋白質の高次構造をNMR法や結晶構造のX線解析法で解明し、がん治療薬を開発するための情報交換を行った。そして偏平上皮癌特定のがんで過剰発現するEGF受容体を介するシグナル伝達系蛋白質の高次構造ならびに転写制御に関わる核内がん原遺伝子産物の高次構造についての情報収集・共同研究で進展があった。また新しい試みであるがんの遺伝子治療に関しても、免疫療法とあわせた手法が米国で試行されておりこの分野にも我が国の医師・研究者を派遣し情報収集した。
