研究概要

近年、がん遺伝子、がん抑制遺伝子の研究が進み、がんを遺伝子ならびにその産物の機能に基づいて理解することが可能になった。それと共に、細胞増殖のためのシグナル伝達機構、細胞周期制御の機構、そして細胞死の分子機構の解明が進んだ。また細胞間相互作用の細胞社会学的研究や細胞表面蛋白質の分子生物学的研究に基づく、がん転移の機構についての知見が集積してきた。一方で、がん関連遺伝子の探索を包含するゲノムプロジェクトの急展開が見られている。また、ウイルス発がんに関してもEBウイルスとヒトがん発症の関連で新しい進展が見られた。このようながんの基礎研究が進んでいる中、遺伝子治療のためのベクター開発や、細胞増殖制御機構に関する知見に基づいた、がんの新しい診断法や治療法の開発が急速に推し進められている。さらには、論理的ながんの予防法を確立するための分子疫学的研究が注目されている。
このような、基礎研究の急激な進展、基礎から臨床研究に向けた情報の発信とそれを受けた臨床応用への試みが期待されている状況で、本国際学術研究では、これらの課題についての研究が先進的に進んでいる米国を中心とした北米大陸に、我が国の第一線の研究者を派遣し、研究室訪問や学会発表による、情報交換、情報収集、共同研究を促進させた。一つには、がん遺伝子産物の機能解析とシグナル伝達・転写調節、がん抑制遺伝子産物と細胞周期調節、細胞死、化学発がんの分子機構、ウイルス発がん、細胞接着とがん転移、genetic instability等の基礎研究分野のうち、急速な展開を見せている研究領域で交流をはかった。また一方で、治療診断のためには、遺伝子治療やがん遺伝子・がん抑制遺伝子産物の分子構造に基づく抗がん剤の設計を重点課題としながら、抗がん剤のスクリーニングや放射線治療、免疫療法に関しても研究者を派遣した。さらにがん予防に向けた分子疫学の領域でも交流を図った。そのために、平成6年度は米国・カナダに17名、平成7年度は米国に19名、平成8年度は米国に15名を派遣し、有効に情報交換を行った。その中からは、共同研究へと進んだ交流もあり、成果をあげつつある。
本学術研究では、文部省科学研究費がん重点研究の総括班からの助言を得ながら、がん研究の基盤を形成する上述のような広範ながん研究を網羅しつつも、いくつかの重点課題を設定した。その一つは、いわゆるがん生物の領域に相当する基礎生物学に近いもので、がん細胞の増殖や細胞間相互作用等の分子機構の急激な展開を見せる研究課題である。二つ目の課題は、物理化学の分野との共同して進められる課題で、シグナル伝達分子や細胞周期制御因子の作用機構・高次構造に基づいて、論理的に新規抗がん剤を設計する試みである。この課題では、がん治療薬開発を目的とした蛋白質のNMR解析、X線結晶構造解析を推進する構造生物学者が分担者に加わった。三つ目は、極めて注目度の高い遺伝子治療法開発に関する研究課題である。レトロウイルスベクター、アデノウイルスベクターの開発に関わる基礎側研究者、臨床医師、免疫学者が参画した。
我が国のがん研究のレベルは近年飛躍的に向上し、世界をリ-ドする立場になってきていると言えよう。しかしながら、上記研究課題を効率良く遂行するためには、今後もがん研究を旺盛に進めている米国等の研究者と交流を深める必要がある。また、ゲノムプロジェクトや発生工学的手法による、がん関連遺伝子研究の進展によって生じる新しい課題をも的確に把握し研究を進める必要があり、そのためにも本国際学術研究が重要な役割を果たしていくと考えられる。
