Gタンパク質による神経細胞イオンチャンネルの調節

研究概要	αβγの3種類のサブユニットよりなるヘテロ3量体GTP結合タンパク質(Gタンパク質)は、細胞外のシグナルを認識する受容体から細胞内2次伝達物質を生成する酵素、イオンチャンネルなどのエフェクター分子に情報を伝えるトランスデューサーとして働いている。そのエフェクター分子のひとつであるGタンパク質共役内向き整流カリウムチャネル(GIRK)は、神経細胞の興奮に対して抑制作用を示すことが知られており、神経活動の基本的分子として働き、神経可塑性など高次神経機能においても重要な役割を果たしていることが示唆されている。本研究の目的は、神経系におけるGIRKの構造とGタンパク質による活性制御機構を明らかにすることである。まず、3種類のGIRK(GIRK1,GIRK2,GIRK4)と、Gタンパク質βγサプユニット(G_<βγ>)の変異体をヒト胎児腎由来293細胞およびラット胎児脳より調整した初代培養神経細胞に一過性に発現させ、免疫沈降法と電気生理学的手法を組み合わせてGIRKの複合体形成およびG_<βγ>との相互作用を検討した。その結果、3種類のGIRKのいずれともβサブユニットを介して相互作用することが明らかとなった。一方、293細胞および神経細胞に外来的にGIRKを発現させて、そのチャンネル活性を測定することが可能であり、かつG_<βγ>の効果を電気生理学的に解析することも可能であることが明らかとなった。今後、分子生物学的手法と電気生理学的手法を組み合わせてGIRKの制御機構を解析することにより、Gタンパク質によるシグナル伝達機構の新たな調節メカニズムが明らかになるのみならず、神経活動におけるGIRKの役割がより鮮明となり、神経疾患や高次神経機能の分子的理解へと繁がることが大いに期待される。
