研究概要

1.化学的研究(国内グループ):癌細胞のPG耐性に関与しているGS-Xポンプタンパク質捕獲のための光親和性標識用プローブを設計し、その合成に成功した。さらに、GS-Xポンプによる輸送機構の検討から腫瘍細胞のドキソルビシン耐性の分子機構を明らかにした。一方、PGの特異的タンパク質誘導作用に直結する細胞内受容体の捕獲・同定のための分子プローブを設計し、その合成法を確立した。具体的には、△^7-PGA_1に構造修飾を施すことで、科学的および生物学的により安定でかつ充分な活性を持つPG類縁体を創製し、これを母核として光親和性標識のためのアジド基と検出のための放射核を組み込んだプローブの合成に成功した。
2.生物学的研究(米国グループ):上記国内グループが合成したGS-Xポンプ機能探索プローブを用い、GS-Xポンプを過剰に発現しているシスプラチン耐性HL-60細胞から調整した形質膜ベシクルでのATP依存性ロイコトリエンC_4輸送阻害実験を行ない、探索プローブとGS-Xポンプとの間に強い相互作用があることを確認した(IC_<50>=0.3μM、Km=110nM)。さらにPG-グルタチオン抱合体輸送阻害実験を行い、同探索プローブが抱合体の輸送を強く阻害することを見出した(IC_<50>=0.5μM)。以上の結果から、探索プローブが、癌細胞の示すPG耐性の克服に向けた新規薬剤設計のリ-ド化合物と成り得ることが明らかになった。現在、国内グループが開発した上記分子プローブを用い、GS-Xポンプタンパク質の捕獲と分子構造の解明、さらにはPGの細胞内受容体の捕獲・同定実験が進行中である。
以上のように研究は所期の目的を達成し、充分な成果を上げることができた。
