γδ型Tリンパ球の生体防御機構における役割

研究概要	BALB/cマウスにSalmonella cholerasuis 31N-1(弱毒株)を腹腔内に投与した。感染6日目で腹腔内および肝臓にγδ型T細胞が増加したが、感染10日目なるとその比率は減少した。感染早期に増加してくるγδ型T細胞をパンニング法でenrichして98%以上の純度のγδ型T細胞を得た。γδ型T細胞はサルモネラ感染マクロファージおよびマイコバクテリア由来のHsp60に反応して、γIFNを産生した。そのVレパートア-はγ鎖はVγ1/Vγ4,δ鎖はVδ6およびVδ5が優位をしめた。次にこのγδ型T細胞のクローニイングをおこなった結果、Vγ1/Vγ6T細胞クローンとVγ4/Vγ6T細胞の2種類がクローン化できた。そのうちγ1/Vγ6T細胞クローンはHsp60に、Vγ4/Vγ6T細胞クローンはBALB/c脾細胞およびMethA腫瘍細胞に反応し、その反応性は抗Hsp60抗体であるML-30にて阻害された。γδ型T細胞のTCRとLFA-1のクロスリンキングによる反応性を検討した。αβ型T細胞は抗TCR抗体と抗LFA-1抗体で強い増殖反応を示したが、γδ型T細胞は抗TCR抗体単独では増殖するものの、抗TCR抗体と抗LFA-1抗体の刺激には全く反応性を示さなかった。DNA解析ではこのシグナルによってアポトーシスが誘導されていることが分かった。In vivoに抗LFA-1抗体を投与するとサルモネラ感染後の肝臓におけるγδ型T細胞の増加が抑制され、菌の増加が認められた。感染症においてγδ型T細胞が何故一過性に増加してその後減少するのか、アポトーシスの感受性で説明されるのかも知れない。
