研究概要

甲状腺刺激ホルモン(TSH)受容体の細胞膜貫通領域を変異させ、Cos-7細胞にトランスフェクションして表現実験を行い、非刺激下、TSH・TSAb(バセドウ病甲状腺刺激抗体)刺激下でのcAMP・イノシトール燐酸産生能とTSH結合能の測定を各変異体についておこなった。その結果、第2細胞内ループの中央部がGsとの相互作用に重要であることが明かとなった。また、Gqとの相互作用については以前報告していた第3細胞内ループの他、第1細胞内ループ全体、第2細胞内ループ、細胞内末尾のN末側と広く分布していることを明かとした。細胞外ループについても同様の検討を行い、第3細胞外ループもcAMP系とイノシトール燐酸系の両方のシグナル伝達に重要であることを明かとした。プランマー病に見いだされたTSH受容体の第6細胞膜貫通ヘリックス部の4種類の点変異がいずれも刺激無しで活性化されている、いわゆるconstitutive activationを起こすものであることを表現実験によって示し、疾患の直接の原因となっていることを証明した。また、糖付加部位のミュータジェネシスにより、TSH受容体のアスパラギンリンクの糖鎖には種差があること明かとした。LH受容体については、家族性男子思春期早発症(FMPP)において新たな遺伝子変異、Met^<571>→I1e、Thr^<577>→I1eを同定し、表現実験によってやはりconstitutive activationを起こしていることを確認した。さらに、家族性のない孤発例で、通常のFMPP患者よりも早期に過度の思春期早発を起こすもので、Asp^<578>→Tyrを同定し、トランスフェクション実験を行ったところ、FMPPの場合の変異よりもはるかに強いcAMP系のconstitutive activationと、FMPPの場合には見られなかったイノシトール燐酸系のconstitutive activationを確認し、病態と極めてよく一致することを見いだした。
