研究概要

我々はまず、ヘルパーT細胞との接触によるB細胞のアポトーシス抑制やクラススイッチ誘導の際に、ヘルパーT細胞上のCD40LとB細胞上のCD40分子の反応によるCD40シグナルの関与を明らかにする目的で、種々のB細胞株を抗CD40抗体やCD40L発現T細胞、精製CD40L分子で刺激した。その結果B細胞株CH12のIg遺伝子のクラススイッチが高頻度で誘導され、またB細胞株WEHI-231の抗原レセプタークロスリンクによるアポトーシスは、ほぼ完全にブロックされた。これらの知見から、T細胞との細胞接触によるB細胞のアポトーシス抑制やIgクラススイッチでCD40/CDL40系が重要な役割を果たすことが明らかとなった。
次に、FDCとB細胞の反応機構を解明するために、我々は株化FDCと種々の株化B細胞を共培養したところ、FDC株がB細胞株WEHI-231の抗原レセプタークロスリンクによるアポトーシス誘導を阻害することを発見した。FDC株はすでにCD40のリガンドとして同定されているCD40L分子発現をせず、またCD40とIgのキメラ分子とも反応しないので他の未知のCD40のリガンドも発現せず、B細胞とCD40を介した反応はできない。またこのB細胞アポトーシスの阻害はLFA-1、ICAM-1、VCAM-1等種々の膜分子に対する抗体では抑制出来なかった。従ってFDC上の未知の分子がB細胞と反応し、アポトーシスを制御しているものと考えられた。また、FDCとB細胞株の共培養では、FDCによるIg遺伝子のクラススイッチ誘導はみられず、FDCによるB細胞アポトーシス抑制機構が、CD40シグナルとは異なっていることが明らかとなった。
