研究概要	我々が発見した世界で唯一のc-kitミュータントラットであるWs/Wsラットを用いて、マスト細胞と好塩基球の分化機構と機能の解析を行うための共同研究を米国、英国、カナダの研究者との間で開始した。我々の研究室で維持しているWs/WsラットはSPFの状態でなく、近交系でもないので、まずWs突然変異遺伝子をDA系に導入し、その後DA-Ws/+ラットをSPF化することにした。5世代DA系にもどし交配したDA-Ws/+ラットを日本SLC(株)に依頼してSPF化し、DA-Ws/Wsラットの生産を開始したが、平成6年度中には十分な量を生産することができなかった。そこで我々の研究室で維持しているWs/WsラットをHarvard大学とMcMaster大学に送り、予備的な共同実験を開始した。Harvard大学へは北村と廣田が出張し、Ws/Wsラットと対照の+/+ラットを用いて、アレルギーショック時の心肺機能を測定する予備実験をGalli、Martinと共同で行ったが、Ws/WsラットがSPFでないため、実験場所に制約を受け、期待した結果は得られていない。ぜひともSPF化したDA-Ws/Wsラットの生産を軌道に乗せる必要があると考えられた。McMaster大学へ送ったWs/Wsラットについても、同大学で独自にSPF化を試みているが、まだ成功していない。一方、Ws/Wsラット由来の培養マスト細胞をEdinburgh大学のMillerにより作製されたラットマスト細胞プロテアーゼ(RMCP)に対する抗体を用いて性格づけようとする試みは、予定通りの成果を得て、共同で論文を出版した。Ws/Wsラットから得た培養マスト細胞は、c-kitレセプターチロシンキナーゼの活性が著明に減少していたが、線維芽細胞との共生培養ではRMCPの表現型が粘膜型から結合組織型に変化したので、c-kitを介したシグナル以外にマスト細胞の表現型に影響しうるシグナルが存在することが示唆された。
