研究概要	本年度の研究の最大の成果は、細胞内のカルシウムイオンの動態を実時間計測する実験装置を開発し、これまでのWhole-cell Voltage Clamp法による膜電流と細胞内カルシウム動体を同時記録する方法を確立した事である。この方法により、これまで間接的な根拠による仮説に留まっていた骨格筋興奮・収縮連関機構に関する我々の考えを証明できた。すなわち、膜内電荷移動(Intramembrane Charge Movement;IMCM)のうちNifedipineで阻害を受ける成分、Nifedipine-sensitive charge movement(I_δ)のみがT-管膜の脱分極シグナルを筋小胞子体(SR)に転送し、SRからのカルシウムイオンの遊離を引き起こす事を証明した。更にこの方法を骨格筋興奮・収縮連関機構の系統発生研究に適応し、これまでに提出していた、骨格筋興奮・収縮連関機構獲得の分子進化が頭索類,Amphioxusと無顎類,Lampreyの間で起こり、それはI_δの発現と一致するという仮説を証明した。膜電位による細胞内カルシウムイオン遊離機構(DICR)はナトリウムスパイクによる膜電位変化に追随し、カルシウムチャネルが開きカルシウムイオンが細胞内に流入する以前に細胞内カルシウムイオン放出を起こさしめる最も速いカルシウム制御機構である。我々はDICRを神経に探った。その結果ザリガニ運動神経細胞にこの機構があるらしい事、それがI_δによって制御されているらしいことを突き止めた。現在、急ぎこの事の証明に取り組んでいる。このことが証明されれば、DICR機構は骨格筋にのみに備わった特異な機構であるのではなく、膜電位を直接利用した細胞内カルシウムイオンの制御という生物の持つ基本的な機能の一つとして広く分布している事になる。
