研究概要

(1)濱、有井、エリスングループ:濱、有井は生理研の1000KV電顕レンズを改良し、優れた像質の傾斜写真を用いて2〜3μmの厚い試料によるコンピュータートモグラフィー(CT)解析を行う体制を整備した。エリスマンのグループは、UCSDの400KV電顕を用いて薄い(1μm)連続切片による連続CT解析を行う体制を整備した。両グループの相互訪問により、星状グリア細胞突起のCT解析の研究を進め、連続CTと単独CTの特性の比較を行っている。(2)濱、有井、大塚グループ:超高圧電子顕微鏡による高分解能三次元定量解析の道を拓くことを目的として、電子顕微鏡画像の直接ディジタル取り込みの研究を行っている。イメージングプレートの高感度性と広い感度域を有効に利用するために、その特性の検討を進め、1000KVでの特性を明らかにすることが出来た。この方法は他の米国グループとの共同研究に利用することが出来る。(3)小坂、Kitaグループ・海馬、嗅球を構成する多様なニューロン群について共焦点レーザー顕微鏡、電顕及び超高圧電顕での解析を進め、海馬においてGABAニューロンにおけるGABA入力が大きく異なることを三次元定量解析により明らかにした。これは海馬におけるリズム形成に重要なGABAニューロン間同期活動の形態的基礎の一つであると考えられる。また海馬モツシ-細胞樹状突起スパインの超高圧CT解析を進めている。(4)村上、ウイルソングループ:村上はテネシー大・ウイルソン研究室を訪問し赤核細胞発生時に見られる樹状突起上のフィロポヂア様の突起三次元解析システムの開発研究を行った。(5)葉原、エリスマングループ:生理学研究所の各種顕微画像システムを用いて、新しく開発した膜電位感受性色素の特性テストを行った。海馬および小脳スライス標本および分離神経細胞による実験の結果、これらの色素が細胞に対する毒性、神経細胞特異性、反応感度などの点で極めて優れた特性を持つことが分かった。
