研究概要

共同研究の最終年度では、それぞれの研究グループが生理研、カルフォルニア大、テネシー大、九大を相互訪問して各プロジェクトの研究を続行、結果の集約、反省を行った。
1)濱、有井、エリスマングループ:生理研超高圧電顕の対物レンズの改良、及びモニターシステムの改善によって連続傾斜画像の質が向上し、またトモグラフィーソフトも改善されたので困難を極めた星状グリア細胞突起の解析が可能となった(結果は1997年アメリカ顕微鏡学会で発表予定)。解析精度を更に上げるために、【.+-。】60度以上の傾斜可能な試料台の開発を検討した。
2)小坂、濱、ウイルソングループ:厚い試料のダイセクター法による定量計測法を確立し、この方法を用いて嗅球、及び海馬に存在する化学的性質を異にする介在ニューロンの定量的な解析に成功した。さらに共焦点レーザー顕微鏡を用い、それぞれの介在ニューロン間の関係を定量的に解析した。主ニューロンについては超高圧電顕を用い、樹状突起刺の形態、及び数の三次元的計測を行った。その結果、特に海馬の背側と腹側で介在細胞のタイプ、及び密度、主細胞での樹状突起刺の形態、及び数に大きな差が存在することが分かった。
村上、濱、ウイルソングループ:今年度は皮質→赤核投射路におけるシナプス形成過程を超高圧電顕による三次元観察法を用いて観察した。バイオサイチンでラベルした皮質→赤核路終末を受ける赤核→脊髄路起始ニューロンの樹状突起上に多数のスパイン状突出物(SLDP)がみられ、これがシナプス形成の初期段階に関与していることを示唆する所見が得られた。
大塚、葉原グループ:大塚は金魚網膜錐体細胞でのオプシン形成、及び外節への出現過程を分子生物学、及び共焦点レーザー顕微鏡法を用いて解析し、オプシンは日没後に形成され、夜間に外節直下に小胞輸送されることを明らかにした。葉原は筋上皮細胞の神経支配をカルシウムイメージング法で明らかにした。
