研究概要

今年度の主要な成果は次の通りである。
(1) 組換えの過程で、塩基配列の相同な二つのDNA分子がどのようにして識別されるかは、最も基本的な問いの一つである。我々はRecA蛋白質・DNA複合体の構造をNMR分光技術を用いて解析し、DNA分子が塩基間スタッキングの代わりにデオキシリボース・塩基間スタッキングをとることを発見し、その構造面の変化から、相同性識別機構の分子モデルを提出した(柴田武彦)。
(2) 組換えの開始で中心的な働きをするMRE11遺伝子を発見し、その蛋白質機能の解析を行った。Mre11は中央部とC-末端にDNA結合部位を持ちDNA二重鎖切断導入にはC-末端の結合部位が必要である。Mre11のヌクレアーゼ活性は、DNA二重鎖切断後の末端のプロセシングに関与することを明らかにした。これらの解析により組換え開始の分子機構の研究に突破口を開いた(小川英行、小川智子、柴田武彦)。
(3) Mre11蛋白質は、更にDNA二重鎖切断端同士の再結合を行い、非相同組換えにも関与することが分かり、相同組換え過程と非相同組換え過程の分岐点が判明した。これは遺伝子ターゲッティング改善の糸口になるかもしれない(小川英行、小川智子、坪内英生)。
(4) Rad51蛋白質の活性が、Rad52とRpalによって大きく促進されることを発見した。その詳細な解析によって真核生物の組換え中間体形成の分子機構解明が大きく加速された(小川智子)。
(5) これらの成果の他に、Mre11蛋白質は、細胞周期制御との関連や、テロメアや繰り返し配列長の維持とも関連あることが明らかになってきている。そしてこれらの成果はまた、X線等によるDNA二重鎖切断の修復研究分野にも大きな影響をあたえつつある。
