研究概要	大船は興奮性アミノ酸レセプターの活性化機構とレセプター蛋白の三次構造の解明に向けて、有効なリガンドの分子設計を進め、本年度は、強力なグルタミン酸アゴニストであるカルボキシシクロプロピルグリシン類(CCG)をリ-ド化合物として、三員環状に置換基を有する化合物の合成に成功し、この方法をさらにカイニン酸タイプのアゴニストの設計に適用し目的とする3'-エチレン-CCGの合成が完成した。この結果、レセプターの放射性ラベル基や光親和性基のついたリガンドの開発の目途がたった。古山はイソプレノイド鎖延長酵素をクローン化し、大腸菌内での大量発現系を確立させ、部位特異的変異導入により、C末端部の特徴的な領域ァと、2個存在するCysの酵素活性発現に対する役割の解明を行った。また、ヘプタプレニル二リン酸合成酵素のクローン化二も成功し、この酵素の大腸菌での発現系を確立した。首藤は蛋白の基本的結合であるアミドあるいはウレイドはNメチル化するとアミド構造はシスとなる。この立体的特性を生かし、芳香環上に複数の水素結合性官能基を持つ、この化合物は予想通りシス配置で存在し、核酸モデルであるジシトシル誘導体と相補的な結合を形成した。この結果はアミド化合物が核酸塩基配列認識分子の構築に有用であることを示している。川はクズ培養細胞から、フラボノイド生合成の最初の鍵酵素であるカルコン合成酵素(CHS)とそれと協奏的に働く還元酵素(CHR)をクローニングし大腸菌での大量発現系の構築に成功している。二つの酵素の反応に於ける役割と変異の導入による反応に必須なアミノ酸の同定、大量の酵素の発現と精製、結晶化により、三次元構造の解明とポリケタイド反応の反応機構の解明を目指す段階にある。また大量の蛋白を得るための各種の発現系について検討し、Aspergillus nidulanceでのフェノール酸化酵素の発現にも成功し、またCHSと同種の反応をするスチルベン合成酵素(STS)についてはバキュオバイラスによる大量発現について検討している。
