抗腫瘍性ペプチドドラスタチン10の活性コンフォメーションの探索

研究概要	ドラスタチン10は強力な抗腫瘍活性を有するペンタペプチドである。その活性コンフォメーションの探索および固定による活性の強化を目的として本研究を進めている。全合成によって得られた天然物とその誘導体の細胞毒性試験をおこなったところ,天然物の強い抗腫瘍性が確認できたと同時に天然物より強い細胞毒性がテトラペプチドで発現することが分かった。以上の知見は活性を保ったままで構造を単純化するための指針を与えており、ターゲットと考えられているチューブリンに結合する活性コンフォメーションをモデル化するうえで重要となることが期待される。また、合成ルート上、低収率であったチアゾールを含むDoe単位の合成法を解決することにより天然物およびその誘導体の大量入手が可能となる。この問題点をフェニルアセトアルデヒドイミンへのチアゾールリチウムの不斉付加反応により解決することを試みた。その結果、再結晶を繰り返すことで光学的に純品な付加体を得ることができた。本反応は脱プロトンのおきやすいイミンに対して適用可能であり、さらに複素環を1段階で導入できるために他の天然物合成にも拡張できると期待している。これらの付加体を組み込んだ誘導体を合成し細胞毒性を調べていく予定である。
