ゴルジ体変異とEGFとLDL受容体の選別輸送の機構

研究概要

ゴルジ領域における細胞の巧妙な選別輸送は、驚嘆に値する。その選別の機構を把握するために我々はこの選別輸送の流れを止めたり、変化させる哺乳類細胞の遺伝子変異株を駆使して小胞体-ゴルジ複合領域の輸送機序を明らかにしたいと考えた。そのために、ゴルジ体ならびに細胞内輸送小胞を標的とするモネンシンならびにブレフェルジンAに対する耐性で選択した体細胞遺伝変異株を対象にして研究を進めた。その結果、
1.モネンシン耐性変異株に特異的に発現している遺伝子群をdifferential display法で単離する仕事を進めている。そはの結果、7-8KbのmRNAサイズを示す1プローブを単離した所、脳に選択的に発現していることが観察された。その全cDNAの単離と機能について現在検索中である。
2.ポリオーマウイルスやv-srcの導入によって、EGF受容体の細胞間輸送が著明に変化することを見出した。その機構として、チュブリンがリソゾームの細胞内分布を変化させることが見出された。更に、H-ras導入によるEGF受容体発現の修飾についてはその分子機序として転写因子AP-1の関与を発見した。
