細胞内アスパラギン酸プロテアーゼの選別と輸送機構

研究概要

カテプシンEはカテプシンDとともに主要な細胞内アスパルティックプロテアーゼである。本酵素の細胞内輸送とソーティング機構については全く解明されていない。本研究ではまずカテプシンEのプロテオリティクプロセッシングと活性化機構を検討するため、ヒト赤血球膜およびラット脾臓から前駆体プロカテプシンEを分解精製し、成熟酵素への変換機構をin vitroで解析した。同時に、ラット脾臓からカテプシンEの遺伝子のクローニングを行い、そのヌクレオチド配列から蛋白質一次構造を決定した。続いて、マウス白血病フレンド細胞およびヒトカテプシンE遺伝子を移入したCHO細胞を用いたパルスチェイス実験によってカテプシンEのプロセシングと細胞内輸送経路を調べ、リソソーム酵素のカテブシンDのそれと対比させた。本研究を通じて得られた結果は以下のとうりである。(1)ヒト赤血球膜およびラット脾臓から分離精製したプロカテプシンEのN末端アミノ酸はそれぞれSer^3およびGln^1であった。(2)両プロカテプシンEとも緩和な酸性条件下で主として分子間反応によって急速に成熟型酵素に変換されることが判った。(3)ラットカテプシンEは19個のアミノ酸残基からなるシグナルペプチド、39個のアミノ酸残基からなるプロペプチドならびに337個のアミノ酸残基からなる成熟ドメインから成っており、1個のNグリコシド糖鎖の結合部位を持つ糖蛋白質であった。(4)成熟赤血球では本酵素は細胞膜の細胞質側に膜結合型の不活性酵素として存在しているが、未分化のフレンド細胞では膜結合型の酵素とともに細胞質型の酵素として存在する。(5)フレンド細胞ではカテプシンEは合成後の比較的早い時期に糖鎖の修飾を受け高マンノース型から複合型糖鎖へ変換するが、プロテオリティックプロセシングは非常に遅いか殆ど起こらないことが判った。(6)CHO細胞ではカテプシンEは高マンノース型糖鎖を維持して複合型糖鎖への変換を示さないが、プロ型から中間体そして微量ではあるが成熟型酵素への変換が見られた。(7)電子顕微鏡を用いた観察から、CHO細胞ではカテプシンEは小胞体およびエンドソームに主として観察され、一部細胞質への局在も見られた。
