シナプス長期増強におけるグルタミン酸受容体の役割

研究概要	1.補助金により購入したパッチクランプ用増幅器により海馬興奮性シナプス応答を記録し、グルタミン酸灌流投与により海馬興奮性シナプス伝達の長期増強(LTP)が誘導されることを確認し、また先に高頻度刺激でLTPを誘導した入力ではグルタミン酸を投与しても増強が見られないことから、グルタミン酸灌流投与による増強が高頻度刺激によるLTPと同一の機構で維持されることを明らかにした。 2.NMDA受容体の開口チャネルブロッカーであるMK-801を用いてシナプス前伝達物質放出確率を計測し、LTP発現に伝達物質放出確率の増大は関与しないことを明らかにした。 3.シナプス後細胞のカルシウムチャネル活性化による細胞内カルシウム濃度上昇によりシナプス後細胞のAMPA受容体の伝達物質に対する感受性が増大することを見いだした。 4.NMDA受容体の活性化によりアデノシンが介在ニューロンより放出され、興奮性シナプス伝達をシナプス前抑制することを明らかにした。 5.錐体細胞内に活性型CaMKIIをホールセルピペットを介して投与し、シナプス伝達に対する効果を検討したが、今までのところシナプス応答の増強は見られなかった。現在は、(1)より安定な活性型酵素を作製する、(2)酵素が樹上突起の中をどのような速度で拡散するかを検討する、(3)細胞体でのアゴニスト応答に対する酵素の作用を検討するという方針で研究を続行中である。 6.AMPA受容体単一チャネル特性に対するCaMKIIの作用を検討するための準備実験として、アウトサイド・アウト・パッチでAMPA投与により開口するチャネルの特性を解析中であり、今後はピペット内灌流により活性型酵素を細胞膜内側に作用させ、単一チャネル特性の変化を検討する予定である。
