HIVのTリンパ球への結合と膜融合に関与するT細胞内プロテアーゼの作用機序

研究概要	エイズウイルス(HIV-1)がヒト由来のCD4陽性細胞に感染して増殖するさいに少なくとも二種類の細胞内プロテアーゼが重要な役割を果している。その1つは感染細胞膜上の蛋白質、トリプターゼTL_2(TL_2)でプロテアーゼ活性を持ちgp120のV3領域と特異的に結合する。両者の結合の見かけ上の解離定数は3.8×10^<-8>MでCD4と比較するとgp120との結合力は弱く、従ってCD4と結合した後にTL_2と結合することが考えられた。また感染細胞膜上のTL_2はいずれもCD4の近傍で、しかもgp120の結合部位の近くに位置していることが判明した。これはCD4に対する抗体の中で、gp120の結合部位をエピトープとする抗体によって細胞膜上のTL_2とその抗体との結合が特異的に阻害されることによる。さらにTL_2と結合したV3ループはウイルスの感染にとって重要と推定される中央部の-GPGARF-モチーフを保存したまま、その近傍の疎水性アミノ酸か塩基性アミノ酸部位で解裂することが明らかになった。第2のプロテアーゼされる過程で、gp160をgp120とgp41へ変換するプロテアーゼである。このプロセシングはウイルスが感染性を示すために必須の過程である。我々はこのプロセシングプロテアーゼ(Viral envelops protein maturase,VEM)が、感染細胞のERとゴルジ膜分画に局在する分子量26±2kDaのCa^<++>非要求性セリン性プロテアーゼであることを明らかにした。さらに本研究でVEMの精製に成功しインヒビター特異性を明らかにした。その結果、VEMは活性中心にSerとその近傍にCysを持つ酵素と推定され、その阻害剤は感染を阻止することが示された。
