tRNAアンチコドン部位のコンホメーションの人工的固定化およびその分子認識能

研究概要	tRNAはアンチコドンループに鋭く曲がったいわゆる"Uターン"構造と呼ばれるベント構造が存在している。この特異な折れ曲がり構造はコドン-アンチコドンの分子認識機構にも関係する重要な構造的特徴である。今回我々は、このベント構造が実際にコドン認識に関与しているか検証する目的でこのUターン構造を化学的に固定したオリゴリボヌクレオチドを合成しその構造解析をおこなった。そこで、まず前年度で合成を完了した環状ウリジル酸2量体が確かにベント構造をとっているか^<31>PNMRにより検討した。その結果、通常の3'-5'リン酸ジエステル結合のリン酸基のケミカルシフトに比べて約1ppm低磁場側にシフトしていることから、強くこのベント構造が示唆された。次にベント構造をtRNAのアンチコドンループ部位に組み込むため、環状ウリジル酸2量体の5'末端リン酸化を行い、ライゲーション反応でtRNAに組み込もうとしたが、ライゲ-スの基質にはならないことがわかった。そこで、5'末端をさらにアデニル化して行ってみたが、やはり目的を果たすことができなかった。そこで、化学的な手法でベント構造をtRNAのアンチコドンループ部位に組み込むため2量体のアミダイト合成ユニットに変換することを検討した。その結果、2量体アミダイトユニットを合成することができた。現在このものを用いて、液相法でUpcUpU3量体の合成を検討している。さらに、核酸の糖部および塩基部位のコンホメーションの人工固定化の研究の一環として、ウリジル酸の塩基5位と5'リン酸基をエチレン鎖で連結したシクロヌクレオチドを合成した。これらの化合物は著しく糖部が3'-endo型に構造が固定化されていることが、詳細なNMRによる構造解析の結果わかった。そこで、さらにこれらのシクロヌクレオチドを骨格とする堅いコンホメーションをもつRNAの化学合成も現在検討中である。
