随意運動制御における大脳から小脳核への入力の機能的役割

研究概要

本研究は、小脳核からの出力、特に最近見いだされた頭頂連合野への投射について解析し、そこから小脳及び大脳運動野ヘ至る出力系について検討した。実験は、ネンブタール麻酔したネコを用いた。頭頂野から運動野へ投射する皮質-皮質投射細胞はII-III層にある錐体細胞で、運動野において数個のブッシュ状の軸索終末を形成しており,小脳電気刺激によって2シナプス性の興奮(EPSP)が生じた。中継核は視床VL核の背内側部であった。中位核・歯状核いずれもが、頭頂葉に投射していた。頭頂葉からの出力細胞のうち橋核投射細胞は小脳刺激によって2シナプス性の興奮を受けていた。頭頂葉の中で上シルヴィウス回と外側回にある細胞では小脳からのEPSPの性質が異なり、前者の細胞では、EPSPの立ち上がりがきわめて緩やかでその潜時が長く、後者の細胞では立ち上がりがより急で潜時も短い。この電気生理学的な特性は、解剖学的解析結果とよく一致し、上シルヴィウス回へは、VL核からI層のみに、きわめて小型の視床皮質投射細胞が投射しているのに対し、外側回では、それに加え、III-IV層により大型の視床皮質投射細胞の投射が確認された。運動野の刺激でこれらのいずれの細胞にも単シナプス性の強力な興奮性入力が観察した。運動野に順行性のトレーサーであるPHA-Lを注入し、この入力が運動野からの皮質-皮質細胞による投射であることを確認した。以上の結果、頭頂葉5野、7野は、小脳からの入力を受け、その出力を運動野及び小脳に送っていること、又、この部位が運動野から強い入力を受けていることが明らかとなった。この部の頭頂葉が運動野から運動司令の情報をfeed-forward的に受けていること、又、体性感覚のfeedback入力と共に小脳からの入力を受けていることは、この部が随意運動制御に重要な役割を果たしていることを示すものである。
