ストレス蛋白発現における活性酵素野役割

研究概要	ストレス蛋白の発現制御の分子細胞生物学的解析が詳細に行われているが、個体レベルでのストレス応答機構の分子論的解析とその制御に関する研究は極めて少ない。ストレス応答には視床下部-下垂体-副腎-血管軸を介する脳-自律神経-循環制御反応系が重要な役割を果たし、これに血管や自律神経系におけるNOラジカルやO_2^-等の活性酸素種が大きく影響する。また、強度のストレスが免疫能を弱体化することが知られている。しかし、いずれもその分子論的実態は不明であった。本研究では、各種の酸化ストレス下におけるSOD、Heme Oxygenase(HO),HSP-70などの遺伝子発現動態を上記Axisにおいて動力学的解析、及び生体の主要抗酸化物質であるグルタチオンの代謝動態の解析を行い、以下の結果を得た。 1)水浸拘束負荷ラットでは肝のグルタチオンが著明に減少して胃潰瘍病変が出現した。この際、胃粘膜のHSP70やHOが一過性に発現増強する現象が見られた。胃粘膜病変はグルタチオンや病巣指向性SODで改善されたが、ストレス蛋白の発現動態は影響されなかった。 2)水浸拘束や低温ストレス負荷ラットでは胸腺リンパ球の著明なApoptosisが誘起されることが判明した。本現象は病巣指向性SODでは影響されず、グルタチオンやN-acetylcysteineで大きく影響された。SOD誘導体は細胞外に分布するが、上記のチオール化合物は細胞内や核内にも分布することから、上記のストレス病態を核内と細胞外の活性酸素代謝との関係でさらに解析することが重要であることが判明した。 3)水深拘束ストレスや低温ストレス負荷ラットでは肝や副腎のグルタチオンが著明に減少することが判明した。現在、GSHおよび関連化合物により生体のチオール代謝を制御し、胸腺の免疫病態とストレス蛋白および活性酸素代謝酵素系の発現制御機構の関係を分子論的に解析しつつある。
