超好熱菌のストレスタンパク質とその応答

研究概要	ストレスタンパク質は高温処理で発現されることから発見された。従来の予想を覆して高熱環境に育つ好熱菌にもストレスタンパク質があり、熱応答と熱ショック応答遺伝子がある事については既に申請者が報告した。本研究はさらに高温で増殖する超好熱環境に耐える超好熱菌を海洋バイオ研究所(帆秋、丸山両研究員)との協同で解析する。そしてそのストレスタンパク質HSP70,60,10相同タンパク質とその遺伝子の構造機能を決定して、極限の熱ショック現象を解明し、超安定な熱ショック蛋白質を得ることを目的としている。海膨や海底温泉から沸騰水またはこれに近い温度で増殖する超好熱細菌を単離して培養した。培養条件は粉末硫黄を要する嫌気的条件による。この硫黄依存性の超好熱古細菌Desulfurococcus strain SY (96℃,東太平洋産),Pyrococcus sp.(104℃,中硫黄島の海底の千鳥温泉産),Thermococcus sp.(96℃,南西諸島小宝島海底温泉産)などからそれぞれ粗DNAを抽出した。超好熱菌のDNAはGCの水素結合を維持出来ない高温で機能するため、強固に保護タンパク質が結合していた。そこで、proteinase K,SDS,Triton-Xで可溶化し、3回のPhenol抽出、RNase処理、クロロフォルム抽出、エタノール沈澱を繰り返した。このDNAに対してHSP70のDNAをプローブとハイブリッドを形成するDNA断片を得た。DNA精製の過程で断片が細分化されたため、再度、DNA処理を続行した。決定された塩基配列はむしろAT含量が高かった。一部の読み枠の構造を決定して現在海洋バイオ研究所より特許申請の予定である。
