研究概要	我々は、タバコからMAPKKKホモログであるNPK1をコードしているcDNAをクローニングし、その構造的、遺伝学的、生化学的・生理学的性質を研究している。NPK1は出芽酵母のMAPKKKホモログであるBCK1の変異を相補できる。このことから、高等植物にもBCK1が関与するようなシグナル伝達系が存在する可能性が考えられる。本年度は以下の4点を明らかにした。 (1)NPK1の機能に迫るために、NPK1プロモタ-とGUSレポーター遺伝子との融合遺伝子を持つ形質転換タバコを用いて、そのタバコ個体における発現部位を詳細に解析した。その結果、NPK1は分裂細胞を含む茎頂と根端で強く発現していることがわかった。この結果は、NPK1が植物細胞の増殖に機能しているという考え方を強く支持するものである。 (2)NPK1抗体による免疫沈澱複合体の解析により、NPK1には実際にプロテインキナーゼの活性があることがわかった。 (3)NPK1cDNAを用いてシロイヌナズナ(Arabidopsis thaliana)のゲノムからホモログをクローニングし塩基配列を調べたところ、この植物には少なくとも4コピーのNPK1ホモログが存在し、キナーゼ領域に関しては互いに80%、全体としては54%のアミノ酸が同一であった。 (4)NPK1の下流にくる因子を得るために、PCRによりタバコからMAPKKホモログをコードするcDNAをクローニングし、それに対応する遺伝子をNPK2と名付けた。NPK2のキナーゼ領域のアミノ酸配列はショウジョウバエのMAPKKホモログであるDsor1と45%同一だった。 (5)NPK1の上流にくる因子を得るために、酵母の実験系を用いて植物のcDNAをスクリーニングした。その結果二種類のポジティブクローンが得られた。現在、塩基配列を決定しているところである。
