研究概要	本研究者はこれまで細胞内情報伝達のメカニズムに興味を持ち、その解析に必要と考えられる細胞質から核への蛋白質の輸送機構の解明を目指してきた。細胞周期を調節するメカニズムの1つとして細胞質から核へのシグナル因子の移行の制御が考えられ、細胞周期の進行に重要な働きをすると考えられるRCC1-Ran/TC4システムに着目して研究を進めてきた。RCC1遺伝子に変異のある温度感受性突然変異細胞株であるtsBN2細胞を用いたこれまでの研究から、主として核内に豊富に存在する低分子量G蛋白質であるRan/TC4が細胞質から核への蛋白質の輸送を制御していることを明らかにしてきた。本年度は、さらにこの研究を押し進め、分子レベルでの詳細な解析を行った結果、以下の事実が明かとなった。 1.培養細胞の細胞質に導入したRan/TC4に対する抗体は、核蛋白質の核への移行を阻害した。 2.tsBN2細胞を非許容温度下で培養して、核蛋白質の輸送活性が低下している状態で、GTP結合型Ran/TC4を細胞質に導入すると、核蛋白質の輸送活性は回復し、この時Ran/TC4自身も核へ移行することがわかった。野生型の細胞の細胞質GDP結合型Ran/TC4を導入すると、核蛋白質の輸送活性は低下し、この場合、ほとんどのRan/TC4は細胞質に留まったままであった。また、同様に、GTPγSを結合させたRan/TC4を細胞質に導入した場合、核蛋白質輸送は完全に阻害された。以上の事実から、核蛋白質輸送は、Ran/TC4のGTP-GDP交換反応を通して制御を受け、核蛋白質輸送に対して、GTP結合型は促進的に、GDP結合型は抑制的に働くことが明かとなった。今後、この制御機構が細胞周期の進行にどのように働くのかを明らかにしていきたい。
