G1/S移行におけるRB蛋白質の役割

研究概要

RB蛋白質の細胞増殖抑制活性は細胞周期特異的なリン酸化で制御されている。しかし、癌化のメカニズムを考える際に特に重要なG1/S移行期においては、サイクリンD、E、Aをそれぞれ含む少なくとも3種類以上のキナーゼがそのリン酸化に関与すると言われながらも、使い分けの機構などが不明であるので、それを解明することを目的とした。そこで、バキュロウィルスベクターを使って、サイクリンD1/Cdk4、サイクリンE/Cdk2とサイクリンA/Cdk2をそれぞれ発現・精製した。これらの精製キナーゼで多くの種類の合成ペプチドのリン酸化を行い、基質特異性の違いを解析した。さらに、RB蛋白質もリン酸化し、リン酸化ペプチドマップを作って、リン酸化部位の違いを解析した。その結果、サイクリンEやA依存性キナーゼによりリン酸化の共通モチーフはS/T-P-X-K/Rであるが、サイクリンD1/Cdk4のそれは明確に異なることがわかった。また、RB蛋白質上にはサイクリンD1/Cdk4特異的なリン酸化部位があることも見い出し、その部位を変異させたRB蛋白質も作製した。さらに、そのリン酸化部位を特異的に認識する抗体も作製できた。したがって、サイクリンEやA依存性キナーゼとサイクリンD1/Cdk4が同じ配列の所をリン酸化するという従来の説は誤りである。我々の実験が成功したのは、それぞれのキナーゼを精製して用いたことにあると思える。RB蛋白質は、サイクリンD1/Cdk4によりリン酸化で、何か特別な活性変化を来すのであろう。サイクリンD1は癌遺伝子にもなるサイクリンであるので、この発見によって、サイクリンD1が細胞癌化に関与するメカニズムの解明が大きく伸展すると期待できる。
