胸腺外分化T細胞の免疫制御と寛容の分子機構について

研究概要	研究目的:これまで、T細胞は胸腺のみで分化、成熟するものと考えられてきたが、1990年に入ってから筆者らが肝類洞で、また他の研究者により腸間上皮で胸腺外T細胞分化が起こることが明らかにされた。特に肝類洞で分化するT細胞(intermediate TCR細胞)は若いマウスやヒトではほとんど活性化されないが、加齢、細胞内寄生性微生物感染、担癌状態で活性化される。胸腺外T細胞分化の特徴は、自己応答性の禁止T細胞クローンが消去されず、かなりのものが、そのまま含まれていることがある。また、胸腺外分化が活性化するときには例外なく胸腺皮質が萎縮し、通常のT細胞分化が抑制される。本研究では、免疫寛容の破綻は胸腺外T細胞の活性化とその末梢でのクローンの拡大としてとらえなければならないという可能性を検討するのを目的としている。研究成果:マウスの肝のみならず、少数ながらintermediate TCR細胞は脾や胸腺にも存在することが明らかとなった。そして、MlsとVβ^+細胞のシステムを使用して、どの臓器でも自己応答性の禁止クローンはintermediate TCR細胞に限局して存在した。特に胸腺にはTCR^-→TCR-dull→TCR-brightのステージがあるが、いずれの分画にも禁止クローンはほとんど存在しなかった。つまり、禁止クローンは、胸腺の主要分化経路ではつくられず、intermediate TCR細胞の分化経路のみでつくられていた。考察と展望:長い間、自己免疫性の禁止クローンは、胸腺内のT細胞分化経路の破綻によって生じるものと考えられてきた。しかし、本研究によりintermediate TCR細胞の分化経路でのみつくられていることが明らかとなった。この現象は、多くの自己免疫疾患や自己免疫様状態を呈してくる慢性GVH病の病因と深く関連しているものと考えられる。本実験では正常マウスを用いたが、免疫抑制剤使用時あるいはX線照射後にも同様のことが言えるかどうかを検討する必要がある。
