研究概要	スーパー抗原性細菌外毒素、例えばTSST-1(toxic shock syndrome toxin-1)やSEA〜SEE(staphylococcal enterotoxin A-E)は感染症の病原性因子であると同時に、特定のTCR Vβ選択的な強力なT細胞活性化作用をもつ。このT細胞活性化によって産生された大量のリンフォカインが疾患の発症に第一義的に関与すると考えられている。スーパー抗原はマウスに投与すると一過性のT細胞活性化のあと深いトレランス(アナジーとクローンの排除)が誘導される。我々は誘導機序の解析を目的として研究を行い、病原因子としての新しいスーパー抗原の探索とトレランスの解析を行い、平成6年度に下記の研究成果を得た。 1.エルシニア感染症の起炎菌より新しいスーパー抗原(Yersinia pseudotuberculosis-derived mitogen,YPM)を発見し、さらにその遺伝子を解析し、DNA配列、アミノ酸配列を決定した。YPMは131個のアミノ酸残基から構成され(分子量14kDa)、HLAクラスII陽性アクセサリー細胞の存在下に TCR Vβ3,9,13を表現するヒトT細胞を活性化する。 2.Staphylococcal enterotoxin A(SEA)注射によるマウスT細胞のトレランスの解析を行い、(1)IL-2産生とSEA応答性T細胞のIL-2レセプター表現の時間的不連続性がT細胞のアナジー誘導に関与すること、(2)アナジーはSEA応答性IL-2R^+アナジーT細胞がIL-2によって免疫記憶タイプ応答性を回復する初期段階、IL-2R^+アナジーT細胞が応答性を回復しない中期、IL-2RアナジーT細胞にアポトーシスが進行する後期段階の3段階に分けることが出来ることを見出した(論文作製中)。 3.ヒト臍帯血T細胞は成人末梢血T細胞は成人末梢血T細胞と異なりアナジーになりやすいことが見出された。
