興奮性アミノ酸による神経細胞死の選択性とその防護に関する研究

研究概要	興奮性アミノ酸による神経細胞死にはグルタミン酸受容体のサブタイプであるイオンチャネル型受容体が中心的な役割を果たしていることが示されている。一方で、代謝調節型グルタミン酸受容体を刺激すると神経細胞死が抑制されることが近年報告されている。同じアゴニストが、作用する受容体の違いにより神経細胞障害性にも防御性にも働くことは極めて興味深く、神経細胞死を研究する上でグルタミン酸作動性ニューロンの役割が極めて重要であることを意味している。本研究では、代謝調節型グルタミン酸受容体アゴニストである(2S,1′R,2′R,3′R)-2-(2,3-dicarboxycyclopropyl)glycine(DCG-IV)を微小浸透圧ポンプを用いて長時間(10時間以上)持続的に脳室内または脊髄くも膜下腔に前投与したあと、カイニン酸、アクロメリン酸をそれぞれ脳室内、脊髄くも膜下腔に投与し、引き起こされる神経細胞死に対するDCG-IVの防護作用を検討した。DCG-IVの前処理により、これらの興奮性アミノ酸による行動変化・神経病理学的変化が用量依存性に著明に軽減した(最大効果は8-27pmol/hで投与したときに得られた)。DCG-IVの前処理のみでは脳室投与では800pmol/h以下、脊髄くも膜下腔投与では80pmol/h以下の投与量ではラットには行動・病理変化はみられなかった。DCG-IVは細胞内セカンドメッセンジャーであるcyclic AMPを抑制する作用を持つ代謝調節型グルタミン酸受容体mGluR2/3に選択的に作用することから、ある種の代謝調節型受容体サブタイプの活性化が神経興奮抑制及び神経防護に関与すると考えられる。本実験から神経細胞死の制御にグルタミン酸作動性シナプスが大きな役割を果たしていることが示唆される。
