研究概要	本年度は次の3の点に重点を置いて実験を行った。 1.狂犬病ウイルスの神経細胞を導く因子が、ウイルスのマトリックスタンパク質であることが推測されていたが、その実態は明らかではなかった。今回、細胞変性の弱い街上毒と細胞変性の強い固定毒との違いがマトリックスタンパク質自身の性質の違いによるものか、あるいはマトリックスタンパク質の発現量の違いによるものかを、両ウイルスのマトリックスタンパク質遺伝子をクローン化し、発現ベクターに組込んで発現させて、その質的な違いの有無を調べることにより、明らかにすることを試みた。実験の結果は、両ウイルスのマトリックスタンパク質の細胞毒性作用はほぼ同じであることを示すものであった。このことは、神経細胞における細胞変性後の違いがマトリックスタンパク質の発現量の違い、あるいはもしあるとするならば、合成後の修飾の違いによるものであることを示唆する。 2.狂犬病ウイルス感染細胞におけるウイルスmRNAの合成量及びマトリックスタンパク質の合成量の調節機構の有無を調べたところ、マトリックスタンパク質に関してのみ、他のタンパク質にはない調節機構が働くことがわかった。即ち、マトリックスタンパク質をコートする遺伝子は、独立したmRMAとしてよりも上流の遺伝子からの"read through"機構により、読み続けられて、多シストロン性のmRNAの下流側に組込まれて転写される方が多く、この仕組みにより、その発現量が他のウイルスタンパク質と比べて、強く制御さていることがわかった。 3.狂犬病ウイルスと同じラブドウイルス科に属し、増殖の仕組みも類似する水痘性口内炎ウイルスのマトリックスタンパク質の働きと、その細胞死の過程を解析したところ、アポトーシスとは異なる細胞死の過程を活性化すること、及びこの過程がプロテインキナーゼ阻害剤の一つKT5720(これはCAMP依存性キナーゼの阻害剤)によって強く抑制されることがわかった。類似のことは狂犬病ウイルスでも起こると思われるので、現在実験を進めている。
