GTP結合蛋白質をはじめとする光情報伝達分子群の多様性と進化

研究概要	交付申請書に記載の研究実施計画に沿って研究を進め、以下の知見を得た。【1】松果体光受容タンパク質の機能解析ニワトリ松果体cDNAライブラリーから新しい光受容タンパク質の遺伝子クローニングに成功した。このタンパク質の機能解析を行うために、単離したcDNAを293EBNA細胞に導入し蛋白質を強制発現させた結果、発現タンパク質は11シス型レチナ-ルと結合し、約470nmの青色の光を最もよく吸収することが明らかになった。そこで、松果体(pineal)のみに発現する光受容蛋白質(opsin)という意味から、この光受容体をピノプシン(pinopsin)と命名した。ピノプシンは松果体の概日リズムを制御していると考えられる。アミノ酸配列をもとに光受容体の分子系統樹を作成したところ、ピノプシンと視物質は同一の祖先遺伝子から進化したと考えられた。さらに、進化の過程で脊椎動物が出現した時点においては、すでにピノプシンの遺伝子が存在していたと推定された。つまり、進化の過程で失っていなければ、下等な魚類を含めて全ての脊椎動物はピノプシンの遺伝子を持っていると考えられた。【2】松果体Gタンパク質の検索ピノプシンが相互作用すると考えられる三量体型Gタンパク質αサブユニットの検索を行った。その結果、PCR法により視細胞のGタンパク質トランスデューシンαサブユニットと類似のタンパク質cDNA断片を松果体から単離することに成功した。現在、全翻訳領域を含むcDNAの単離を試みている。【3】未知の光受容体の検索松果体とは別に、鳥類の脳深部(視床下部)には、季節性繁殖や概日リズムの光同調を制御する脳内光受容体が存在すると考えられている。この光受容体はピノプシンのホモログである可能性があるので、その遺伝子クローニングを目的として、現在、ハト外側中隔部のcDNAライブラリーを作製している。
