ミトコンドリア遺伝子のヒト種内変異の解析による分子進化機構の研究

研究概要

ヒトのミトコンドリア遺伝子(mtDNA)は、1)組み換えなどの複雑な修復を受けず進化機構が単純である、2)コピー数が大きいため遺伝子増幅が容易でPCR産物の塩基配列を直接決定できる、3)種内変異の頻度が高く大量のデータを集積できる、などの特徴がある。研究代表者は43例の個体のmtDNA(16,569bp)の全塩基配列を決定し約70万塩基対のデータを解析した。蛋白をコードする13種の遺伝子(11,320bp)の295箇所のサイトにおいて塩基置換を観察した。
mtDNAのアミノ酸の変化に影響しない四重縮退部位(4-fold degenerate sites)に注目して解析を行った結果、L鎖においてG→AおよびT→C塩基転位の頻度が高かった。すなわちL鎖上のGおよびTは変異し易いが、L鎖上のCおよびA(H鎖上のGおよびT)は変異し難い。つまり変異が鎖に対して非対称的に蓄積していた。この現象は、mtDNAの複製において親H鎖が一本鎖状態に置かれることと、一本鎖DNAでの脱アミノ反応の速度が二本鎖DNAでの速度よりはるかに高いことに関連があろう。一本鎖状態に置かれた親H鎖上のC残基が脱アミノされuracil(U)となり、これを鋳型として娘L鎖が合成される際にAが対合することによって、G→A塩基転位が生じると推定される。また親H鎖上のA残基が脱アミノされhypoxanthineとなり、これが鋳型となって娘L鎖が合成される際にTではなくCが対合し、結局T→C塩基転位を生じるのであろう。これに対して、親H鎖上のG残基が脱アミノされxanthineとなっても、xanthineはCと対合しやすいために、L鎖上のCは変異を免れると推定された。このようにmtDNAの両鎖の複製時における非対称性はmtDNAの突然変異発生の出現頻度に大きな影響を及ぼしていると結論された。
