心筋症ハムスターの遺伝的地図作成と原因遺伝子探索

研究概要	心筋症ハムスターは、ヒト特発性心筋症のモデル動物として重要であるが、常染色体性の単一遺伝形式にて100%の浸透率をもって発症するにも拘わらず、DNAマーカーが存在しないため遺伝的解析が遅れている。今回、心筋症の原因遺伝子座位を同定するために、シリアンハムスターの全染色体地図の作成を目的とした。主な戦略は、restriction landmark genomic scanning(RLGS)を用いて、RLGS spot mapping法により全ゲノムマップを作成し、その中に、心筋症の表現形質を支配する遺伝子をマップする。まず、RLGSゲルの大量生産系(RLGSVer.1.8)を確立した。RLGSVer.1.8は、RLGS prototype(RLGS Ver.1.0)に比してラベリングの反応系の縮小化を行ない、フェノール抽出によるバッファー系の置換の必要性をなくしたため高速にRLGSパターンの生産ができるようにデザインしてある。予備実験の結果、ACNがBio14.6と多型がもっとも多いと考えられたので、Bio14.6とACN系の戻し交配家系60匹を対象とした。制限酵素の組み合せをNotI-PvuII-PstI,NotI-PstI-PvuII,NotI-EcoRV-MboI,BssHII-PvuII-PstI,BssHII-PstI-PvuIIの5種類用い、Bio14.6とACN、およびそのあいだに生まれたF1のRLGSパターンを作成した。約500個の多型マーカーを検出し、戻し交配によって得られた個体についておのおのRLGSパターンを作成後、その伝達を集計した。連鎖解析にはMap Managerを用い、組み替えの頻度が最小限となるようにした、こうしてできた遺伝地図は、全長約1300cMであり、全ゲノムの90%以上カバーすると考えられる。また、27個のリンケージグループからなり、表現形質は、そのうち1つのリンケージグループの上にマップされた。 [結論]RLGS法そのものRLGS spot mapping法は、我々の開発したオリジナルな方法であり、これをハムスター亜種間の多型解析に応用し、全染色体地図を作成した。これによる心筋症ハムスターの遺伝学的解析の結果、原因遺伝子のマップされるリンケージグループを同定した。
