心筋におけるナトリウム利尿ペプチド/cGMP系の意義に関する分子生物学的研究

研究概要	ナトリウム利尿ペプチドファミリーはAtrial natriuretic peptide(ANP),Brain natriuretic peptide(BNP),C-type natriuretic peptide(CNP)から構成され、膜型グアニル酸シクラーゼそのものであるナトリウム利尿ペプチド受容体を介しその生物活性を発現する。本研究は、ANP,BNPの産生臓器である心臓そのものに注目して、"心臓組織内ナトリウム利尿ペプチド系"の心筋代謝、機能調節における臨床的意義を解明することをその目的とする。今回、ラット培養心筋細胞肥大モデルを確立し、ナトリウム利尿ペプチドの遺伝子発現調節を検討し、更にサイクリックGMPカスケードのシグナルトランスデューサーであるcGMP-dependent protein kinase(Gキナーゼ)について、ヒトGキナーゼサブタイプIαcDNAのクローニングを行った。 Wistar系ラット新生仔の心室組織を用い、培養心室筋細胞を調整した。培養心室筋細胞はEndothelin-1(ET-1,10^<-8>M)添加後24時間よりcell sizeの増大、Myosin Light Chain-2遺伝子の発現亢進を特徴とする細胞肥大を示した。この培養心筋細胞肥大モデルにおいて、心筋細胞からのANP及びBNPの分泌は増加し、更に、心筋細胞そのものにおけるサイクリックGMP産生も有意に増加した。ET-1刺激により、ANPmnRNA濃度は数時間後より徐々に増加したが、BNPmRNA濃度の増加はET-1添加後15分より生じ、BNP遺伝子はimmediate-early geneであるc-fos遺伝子と同等の極めて迅速な発現亢進を示した。BNP遺伝子の迅速な発現亢進は、PAT及びPhenylephrine添加によっても認められ、Protein Kinase C(PKC)阻害剤H7やカルフォスチンCにより抑制された。更に、AMD添加によるRNA合成阻害後、培養細胞内のmRNA半減期について検討した結果、BNPmRNAの半減期は約4時間と短く、ANPmRNAの半減期の3分の1以下であった。最近、心筋及び血管平滑筋において、Gキナーゼはイノシトール3リン酸産生やCa^<2+>-ATPase、Ca^<2+>依存性K^+チャンネル等の活性調節に関与し、細胞内Ca^<2+>濃度を低下させることが報告され、cGMPカスケードの主要なメディエーターであることが明らかとなってきた。そこで今回、グアニル酸シクラーゼ/Gキナーゼ連関の心筋分化と機能調節における意義を明らかにするため、ヒトGキナーゼIαcDNA全長のクローニングを行った。得られたcDNAは全長2213bpより成り、671個のアミノ酸より構成されていると考えられた。これらの成績より、培養心筋細胞肥大モデルにおいて、ANP、BNPが明らかに異なる遺伝子発現調節を受けていることが証明され、これらナトリウム利尿ペプチドが心筋細胞そのものに作用しその機能調節に関与している可能性が示された。更に、ナトリウム利尿ペプチド/サイクリックGMP/Gカスケードの意義の解析が分子レベルで可能となった。
