細胞質分裂における低分子量G蛋白質Rhoの機能と作用機構

研究概要	私共の研究室では、低分子量G蛋白質Rhoがアクトミオシン系を介して細胞形態や細胞運動、細胞膜のラッフリング、平滑筋の収縮、細胞質分裂を制御していることを明らかにしている。そこで、本年度の本研究では、細胞質分裂におけるRhoの活性化機構と作用機構を明らかにする目的で、Swiss3T3細胞や出芽酵母ににおける細胞質分裂時のRhoの細胞内局在を免疫蛍光染色法により検討した。Swiss3T3細胞や出芽酵母では、Rhoは細胞分裂後期までは細胞質に存在したが、細胞分裂終期に入り細胞質分裂が始まると形成された収縮環に濃縮して認められた。さらに、Swiss3T3細胞においては、Rhoは細胞質分裂時にはERMファミリーのメンバー(Ezrin、Radixin、Moesin)やCD44と共に収縮環に局在していることが明らかとなった。ERMファミリーのメンバーはアクチンフィラメントが細胞膜と結合する部位に存在してアクチンフィラメントと細胞膜との結合を制御していると考えられているが、最近、細胞膜上のERMファミリーの結合蛋白質はCD44であることが明らかになっている。したがって、分裂細胞では、Rhoは細胞質から収縮環にトランスロケーションして、ERMファミリー-CD44系を介してアクチンフィラメントと細胞膜との結合を制御することにより、細胞質分裂を制御していると考えられる。以上、Rhoによる細胞質分裂の制御機構がかなり明らかになり、本年度の研究目的はほぼ達成できたと考えられる。
