研究概要	GM-CSFレセプターβcはBox1に結合するJAK2を介してβ鎖のチロシン残基をリン酸化する。新井らはその結果Ras-Raf MAPK経路の活性化とc-fosとegr遺伝子の活性化を誘導することを示した。さらにヒトGM-CSFレセプタートランスジェニックマウスはヒトGM-CSFの投与により赤血球NK細胞の産生を促進することを示した。宮島らはIL-3により誘導される初期応答遺伝子として同定したオンコスタチンM(OSM)の遺伝子発現はSTAT5シグナル経路により制御されること,マウスOSMが胎児のAGM領域での初期造血を強く促進することを明らかにした。高津らはIL-5受容体α鎖遺伝子欠損マウスにおいては、B-1細胞の生成の異常と誘導的な好酸球生成がみられないこと、広東住血線虫感染で虫体の排除が遅延することを明らかにした。また、抗ヒトIL-5Rα鎖抗体を用いて免疫沈降により、IL-5Rα鎖とβ鎖はIL-5刺激により初めて会合すること、α鎖にJAK2がβcにJAK1が構成的に会合していることを明らかにした。長田らはG-CSFは好中球の前駆細胞に作用し、増殖と分化を誘導する。その際、転写因子STAT3がリン酸化、活性化される。STAT3のdominant-negative変異体を用いた解析の結果、STAT3は、G-CSFによる増殖のシグナルに関与していないこと、分化のシグナル伝達系にはSTAT3を介するルートと介さないルートが存在することが示された。松島らは造血調節機能をもつMIP-1αに対するレセプター、CCR1への抗体を作成し、その動態を検討した。その結果、大半のヒトCD34陽性細胞がCCR1を発現しており、さらにMIP-1αは主にCCR1を介してBFU-Eコロニー形成を抑制することが明らかとなった。冨田らはヒトLIFレセプター遺伝子は全長70kb以上あり、20個のエキソンから構成されることを明らかにした。また、ヒト絨毛癌細胞の培養液中に可溶性LIFレセプターを検出た。レセプター遺伝子のイントロンを含んだmRNAから産生されると推察された。溝口らは、巨核球の分化のモデル系を用い、NF-E2が巨核球の多倍体化および血小板糖蛋白の発現に不可欠な転写因子である可能性を示した。また、インドロカルバゾール系化合物、KT6352がMeg-POT活性をもち、血小板増加作用があることを示した。杉山らは白血病細胞では、WT1遺伝子が異常に高発現しており、また、WT1アンチセンスオリゴマーにより増殖が抑制されることなどを見い出した。本吉らは、M-CSFがヒト単球のIL-8産生を促進することによって好中球機能を活性化すること、M-CSF依存性のヒト単球の増殖と分化をIL-10が増幅することを見い出した。
