研究概要	造血幹細胞の定着・増殖・分化・死のシグナルについて、造血幹細胞あるいはその子孫細胞を用いて研究を行い、以下のような結果を得た。 (1)北村はマスト細胞の前駆細胞の組織内への侵入機構を調べる実験系を組み立てた。小腸粘膜にマスト細胞の著明な増加を引きおこす寄生虫Nippostrongylus brasiliensisを感染させると、末梢血中のマスト細胞前駆細胞が小腸粘膜内に侵入した。 (2)西川はIII型レセプター・チロシンキナーゼであるPDGFレセプターα、c-kit、flk1、c-fms、flt2/flt3に対する単クローン抗体を開発し、骨髄中のCHU-Mの増殖には、c-fmsではなくc-kitが関与することを示した。 (3)帯刀はc-Mycがどのような遺伝子を標的としているのかを解析できる実験系をMEL細胞株を用いて制作した。 (4)太田は血液細胞の増殖・分化に伴う細胞周期調節遺伝子cdc2の発現とその調節機構について調べ、cdc2のmRNAは未分化な前駆細胞では強く発現しているものの分化に伴って発現が低下することを示した。 (5)辻本はbcl-2遺伝子を完全にノックアウトした。bcl-2ノックアウトマウスでは成長するに伴いpreB細胞、proB細胞を完全に欠損した。 (6)中井はマウス骨髄性白血病M1細胞株がIL-6の添加によりマクロファージに分化する際、核が偏在化する現象にビメンチン関与することを示唆する結果を得ているが、今年度はビメンチン遺伝子の転写について調べ、ビメンチン遺伝子上流のCCAAT配列が転写活性を正に調節していることを示した。 (7)平井は慢性骨髄性の急性転化時と骨髄異形成症候群の白血病化に伴ってみられる染色体転座の意義を分子レベルで追求し、2つの転写因子の結合したAML1/Ev-1モメラ蛋白が産出されることを明らかにした。 (8)井川はエリスロポイエチン・レセプターからのシグナル伝達について調べ、エリスロポイエチンの刺激後、レセプターと会合する2種類の蛋白質を得た。
