造血幹細胞制御と臨床応用に向けての基礎研究

研究概要	本研究組織が発足してから2年間が経過し、本年度はその間の研究経過についてのヒアリングを受けた。このための資料は代表者の北村が中心となって集め、須田と新井の助力を得て取りまとめ、総括班員には別個に承認を得た。本研究の実施にあたっては、1)造血幹細胞の単離と増幅、2)造血幹細胞の定着・増殖・分化・死のシグナル、3)造血調節因子、という3つの計画班と各々の計画班に相応する公募研究から研究組織を構成した。幸いにして本研究領域は質の高い公募研究を採用することができ、計画研究も順調に進展したので、当初予想したよりも速く研究が進んだと考えられる。現在までの2年間に、(1)1個の造血幹細胞により致死量の放射線照射を受けたマウスを救命できるほどに造血幹細胞の精製の技術が進歩した。(2)造血幹細胞に発現しているいくつかの受容体型チロシンキナーゼが見つかった。(3)c-kit受容体型チロシンキナーゼの活性化突然変異が見つかり、このうちの1つの突然変異はヒトの造血器腫瘍でも発見された。(4)赤血球、顆粒球、マスト細胞の分化に重要な役割をはたす転写因子の研究が進んだ。(5)エリスロポエチン、G-CSF、GM-CSF、IL-3、IL-5などの造血調節因子の受容体からのシグナル伝達の経路がかなり明らかになった。本研究組織から論文も順調に出版されており、impact factorが5以上の雑誌に掲載された論文のみでも200を超えている。班会議は3班合同で11月10日に開いたが、前日に造血幹細胞シンポジウムを行い、このシンポジウムでの発表は班会議での発表とみなした。班員以外の聴衆も多く、活発な討論が行なわれ、本領域への関心が高いことが示された。また班会議の際に造血幹細胞への遺伝子導入に関する新しい技術を開発した企業の方にもセミナーを行なっていただき好評であった。
