研究概要	「造血幹細胞制御と臨床応用に向けての基礎研究」は平成6年度から平成9年度の4年間の研究期間をおわり多大の成果を上げることができた。本研究は(1)造血幹細胞の単離と増幅、(2)造血幹細胞の定着・増殖・分化・死のシグナル、(3)造血調節因子という3個の計画班を設けて行なわれたが、3班の間の連携は強く、各研究者の間でも密接な共同研究が行なわれた。総括班として合同班会議を大阪で行なったが、計画班員の研究のこの4年間の進展はめざましく、当初考えていたよりも多くの成果が得られた。また公募に応じた研究者の成果もすばらしく計画班員の研究を補完した。本年度に行なわれた各計画研究の特記すべき成果を述べると、(1)の「造血幹細胞の単離と増幅」では造血幹細胞の発生についての研究に進展がみられ、TIE、TEK受容体が造血幹細胞の発生する場所であるAorta-Gonads-Mesonepheros (AGM)領域に発現していることがわかった。造血幹細胞と血管内皮細胞の共通の祖先と考えられるhemangioblastの分離を抗TEK抗体を用いて行える可能性がある。TEKのリガンドであるangiopoietinの造血幹細胞に対する作用も調べられた。angiopoietinは造血幹細胞の増殖を誘導しないが、細胞接着機能を修飾した。(2)の「造血幹細胞の定着・増殖・分化・死のシグナル」ではマスト細胞の分化に重要な役割をはたす転写因子MITFの解析が進み、MITFによりその転写に影響をうける蛋白が数多く同定された。(3)の「造血調節因子」ではGM-CSF受容体からのシグナル伝達の過程が明らかにされた。またGM-CSFがその受容体に結合することにより活性化されたるStat5の標的遺伝子を探索した際にマウスのoncostatin M (OSM)の遺伝子がクローニングされたが、マウスではOSMとLIFは受容体を共用していないことがわかった。
