同種骨髄移植後の造血系・免疫系細胞の再建と免疫寛容獲得におよぼすGVHの影響

研究概要	骨髄移植後、骨髄幹細胞より成熟T細胞へ分化する過程で、ホスト抗原反応性T細胞はnegative selectionによってクローン消去を受ける。抗原として内因性スーパー抗原の一つ、Mls-l^aに注目し、Mls-l^a反応性T細胞そのクローン消去のメカニズムについて解析した。その結果Mls-l^aは、ホストの放射線抵抗性T細胞によって産生されることが判明した。さらに産生されたMls-l^a産物は、ドナー幹細胞由来のMHCクラスII陽性細胞(マクロファージ、樹状細胞)に移行され、その後Mls-l^a-クラスII複合体が、Vβ6^+またはVβ8.1^+のMls-l^a反応性T細胞を胸腺髄質内において消去することを明らかにした。しかし、マイナ-GVHRの存在下では、ドナー成熟CD8^+T細胞によって、トレロジェンを産生するホスト放射線抵抗性T細胞が完全に排除されるため、Vβ6^+またはVβ8.1^+T細胞のクローン消去が生じなかった。この事実は、マイナ-GVHRによる発癌性ウイルス産物を発現する細胞の排除(graft versus leukemia)への道を拓くもので、今後白血病の治療として骨髄移植を行う場合に、基礎データとなると考えられた。トレロジェン(Mls-l^a)産生細胞は、主としてCD8^+T細胞で、CD44^+CD8^+ T細胞は中枢核、末梢性いずれのクローン消去も誘導したが、CD44^-CD8^+ T細胞は胸腺内のクローン消去を生じなかった。この結果からCD44分子は、T細胞が胸腺に入る際のレセプターとなっていることが示唆された。最後に、胸腺内にNK1.1^+TCR α/β^+細胞が同定された。これらはCD4^+8^+未熟型胸腺細胞をFasを介して殺すことが判明した。現在NK1.1^+TCR α/β^+ T細胞によるGVHRの誘導と制御メカニズムを解析中である。
