研究概要

(1)放射線抵抗性の異常な幹細胞を有するMRL/lprマウスでは、9.5Gyの^<60>Coを照射し、正常マウスの骨髄を移植しても、3カ月以降になると、MRL/lprマウス由来の残存していた幹細胞より、異常なT細胞,B細胞,マクロファージが出現し、自己免疫疾患が再発して来る(Proc.Natl.Acad.Sci.86:3306-3310,1989)。そこで、pluripotent hemopoietic stem cells(PHSC)を精製する方法を開発し、PHSCの分化、増殖をin vitroで解析した結果、PHSCが、増殖分化するためには、骨髄の微小環境構成細胞(stromal cells)が必須であること、さらに、骨髄移植(BMT)の際、PHSCとstromal cellsとのMHCが一致している方が、PHSCが増殖し易いことが判明した。そこで、BMTの際、PHSCだけでなく、donor由来のstromal cellsの補充が必要と考え、骨を移植した。骨と骨髄を同時に移植したMRL/lprマウスでは、1年近く経過しても100%の生存率を示し、stromal cells移植の重要性が明らかになった(J.Immunol.152:3119-3127,1994)。(2)最近、慢性腎炎の中でも治療抵抗性のfocal segmental glomerular sclerosisのモデルマウスが開発され、このマウスを用いて慢性腎炎もstem cell disorderであることを実証することができた(J.Exp.Med.179:1053-1058,1994)。(3)骨髄の微小環境を構成しているstromal cellsがPHSCsを呼び寄せる因子を分泌していることを見い出した(Blood83:964-971,1994)。(4)アロのPHSCsが肝臓にtrapされることにより、アロの抗原特異的に免疫抑制が起こることを見い出した(Eur.J.Immunol.24:1558-1565,1994)。(5)多能性造血幹細胞(PHSC)を精製する新しい方法を発見した(Proc.Natl.Acad.Sci.in.press)。この方法では、4コのP-HSCにより280日間の骨髄再建が可能である。
