研究概要	研究成果のまとめ膵多段階発癌におけるDNA異常の解析と発癌機構および発癌抑制機構について分子レベルから生体レベルまで総合的に研究を行い以下の成果を得た。ハムスター膵発癌過程においてK-ras遺伝子変異は発癌早期に生じる遺伝子異常であること、p53遺伝子異常は膵癌の進展とともに生じる発癌後期の異常であることが示された。また、膵液により膵発癌性ニトロサミンが特異的に活性化されることより、この発癌系において膵液の関与の重要性が指摘された。膵発癌抑制物質として、DNAアルキル化の抑制剤とされているphenethyl isothiocyanateがBOPによるハムスター膵発癌を抑制することが見いだされた。生物学的に、ハムスター膵管病変は末梢膵管に発生したものの方が中枢膵管に発生したものより増殖能の高いことが示された。イヌにおいても膵発癌実験系がほぼ確立された。ヒト膵癌においては、その発生と進展にK-ras遺伝子やp53遺伝子以外にもp16遺伝子などのDNA異常の関与が示唆され、また膵癌細胞株において未知領域のDNAに異常があることが見いだされ、この領域の遺伝子の検索が進められている。膵管癌以外の組織型の腫瘍のDNA異常についても成果は蓄積されつつある。また、ヒト膵癌の肝転移に関与すると考えられる血管内皮細胞の開裂を促進する新たな糖蛋白の存在が示唆された。膵癌の存在診断において膵液よりのK-ras遺伝子変異の検出は早期診断には有効といえないことが明らかにされた。これら本年度の成果は膵癌克服のための貴重な情報となりうるものである。
