転写因子PEBP2と白血病発症の機序

研究概要

転写因子PEBP2は2つのサブユニット、α及びβで構成されている。2つのサブユニットをコードする遺伝子のそれぞれがプロトがん遺伝子であることが知られている。PEBP2の構造と機能を解明し、ヒト急性骨髄性白血病の発症機序を明らかにする目的で解析を続け以下のような結果を得た。
1. PEBP2は単独では転写を活性化できない。PEBP2による転写活性化はcontext dependentである。
2. PEBP2α及びβサブユニットのどちらにも転写活性化ドメインはない。
3. TCRβエンハンサー全体の活性にはTβ3及びTβ4に存在するPEBP2及びEts結合部位の少なくとも一つずつが必須である。
4. PEBP2とEtsとは機能的にも共同作用がある。
5. αサブユニットとEts-1はDNA非存在下で直接結合する。
6.キメラ蛋白AML1/ETO(MTG8)及びβ/SMMHCはいずれもcontext dependentの転写活性化の起こる条件下で完全長のαサブユニットによる転写活性化を干渉する。
7. PEBP2はGM-CSFの発現制御に関与する。
8. PEBP2αB1のヒトホモログ(完全長のAML1蛋白)をコードするcDNAを単離した。
