B型肝炎ウイルスX蛋白質と結合する肝細胞内プロテアーゼの機能変化と発癌機序の解析

研究概要	B型肝炎ウイルスのX蛋白質は、感染した肝細胞内で細胞増殖モジュレーターとして作用して、肝癌の発症に関与していると考えられているがその作用機序については不明である。これまで、X蛋白質がその分子内に含まれる2ケ所のKunitz型プロテアーゼインヒビター類似構造領域を介して、肝細胞内のいずれかのプロテアーゼと結合する可能性が推定されてきた。そこで肝細胞に含まれるいくつかのプロテアーゼの中で、我々の見出した細胞膜付着型と細胞質局在型の2つの局在型を持つセリン性プロテアーゼのTryptaseTL_2を肝細胞より精製し、X蛋白質との相互作用を検討した。本年度はHep G2細胞の細胞質分画より精製したTryptase TL_2を用いリコンビナントX蛋白との相互作用をBIAcoreを用いて検討した。両者の結合の見かけ上の解離定数はKd=6.3×10^<-6>Mで比較的弱い結合を示した。しかしTryptase TL_2のプロテアーゼとしての活性をX蛋白質は濃度依存的に抑制し、Tryptase TL_2とX蛋白質との結合はTryptase TL_2の活性抑制と相関することが示唆された。なお、低分子ペプチド性基質を用いたTryptase TL_2活性のX蛋白質による阻害は、高分子蛋白質(例えばHIV gp120)水解活性のX蛋白による阻害に比して弱い阻害を示し、抑制効果は約1/10であった。このことは、肝細胞内でTryptase TL_2が高次構造を持つある特定の基質を限定分解するさいに、X蛋白質は低濃度で特異的にこの水解を阻止することを示唆する。これまでにX蛋白質は肝細胞内でMAPキナーゼを活性化することが知られている。そこでMAPキナーゼ活性を上昇させる細胞内シグナル伝達系因子が、Tryptase TL_2の細胞内での水解基質になりうるか否か、水解によりその因子は活性化されるか失活するか、これをX蛋白質が阻止するか否か、さらに結果として細胞内のシグナル伝達系がどのように変化するか今後検討を進める。
