抗脂肪肝因子欠乏食によるラット肝発癌機構へのアラキドン酸カスケードの関与機構解析

研究概要

抗脂肪肝因子欠乏食であるコリン欠乏アミノ酸(CDAA)食によるラット肝硬変及び肝発癌誘発機構へのアラキドン酸カスケードの関与機構を、種々のアラキドン酸カスケード阻害剤を用い、肝細胞障害の誘発、癌遺伝子発現、活性酸素産生の観点から検索し、以下の結果を得た。
1.Cyclooxygenase(CYO)阻害剤であるaspirinは、CDAA食によるラット肝細胞壊死の誘発を抑制する事が判明し、CDAA食によるラット肝硬変及び肝発癌の誘発に対するaspirinの抑制機構には、肝細胞壊死の抑制が関与している事が示唆された。
2.Aspirinは、CDAA食投与7日の早朝に誘発されるc-myc発現をむしろ亢進せしめる事が判明した。Aspirinによるc-myc発現の亢進と肝細胞壊死の抑制がどのように関与しているのか、今後更に検索する必要がある。
3.CDAA食投与12週におけるc-myc,c-jun,c-Ha-rasの発現に対し、aspirinは有意な抑制効果を示さなかった。CDAA食によるラット肝発癌機構へのc-myc発現の関与が指摘されているが、aspirinによりその発現が抑制される癌遺伝子の検索が今後必要とされる。
4.CYO阻害剤であるindomethacin(INDM)は、aspirinと異なりCDAA食によるラット肝硬変及び肝発癌の誘発に対し有意な抑制効果を示さなかった。Lipoxygenase阻害剤であるnordihydroguaiaretic acid(NDGA)は、quercetinと異なり、CDAA食によるラット肝発癌を軽度から中等度抑制するも、肝硬変は抑制しなかった。活性酸素産生の指標である8-hydroxydeoxyguanosine(8-OHdG)の誘発に対しては、INDMは抑制傾向を示すもNDGAは有意な抑制効果を示さなかった。AspirinとINDMとの作用の相違は重要であり、今後さらに検索する必要がある。
