RLGS法による癌DNA変化のスクリーニングと癌抑制遺伝子単離システムの開発

研究概要	従来のRLGS法を根本的に改良したRLGS Ver.1.8の開発とその応用-マウスがん組織のDNA変化の系統的検出システムの開発を行った。RLGS Ver.1.8は、RLGS prototype(RLGS VEr.1.0)に比してラベリングの反応系の縮小化を行ない、フェノール抽出によるバッファー系の置換の必要性をなくしたため高速にRLGSパターンの生産ができるようにデザインしてある。マウスがん組織のDNAは、易発癌性のSV40T-antigenのトランスジェニックのC57BL/6とM.spretusのF1の肝臓ガン組織を用いた。コントロールとして正常の肝臓組織を用いた。マウスにおいてNotI-PvuII-PstIの制限酵素をセットを用いて、我々がすでに染色体上にマッピングした323個に加えて、今回新たに229個のspotsをマッピングした(計552個)。これらのspotsは、一画面上で検出でき、一枚のフィルムにより全染色体上の552個の座位をスキャニングできる。また非多型spotではLOHの領域でかりにがん細胞が100%としてもの濃度が半分になる。しかし、マップしたspotsは、多型spotであるためspotの出現消失で識別できる。このマーカーを用いて、マウスの肝がん組織において、4番等に染色体高率に共通する欠失を検出することができた。 (結果)RLGSVer.1.8は従来の原法に比して約10倍のコストダウンとスペースの縮小化を可能とし、一日の処理能力も約10倍にあげることができた。RLGSVer.1.8を用いて1枚のゲルでマウスの全染色体を552個のspotsでスクリーニングできる地図を作成した。全ての生物種にも有効に適用可能なRLGS高速ゲノムマッピング法をさらに加速化した。RLGSspot mapping法は他の全ての生物種にも有効に適用可能であり、さらにC57BL/6とM.spretusのF1のがんDNAのRLGS画像を用いて、たった1枚のゲルで552個のmapped lociを系統的にloss of heterozygosity(LOH)および増幅(amplification)で検出することができた。以上のようにマウスのガン組織を用いて高速かつ効率的にLOHを検出することのできる系を確立し、これを用いると高速に全ゲノムのLOH amplification mapをマウスで作成できる。
