研究概要

オカダ酸クラス発がんプロモーターの研究はいろいろな臓器に共通する一つの発がんプロモーション機構を明確にした。この機構をヒト発がんプロモーションに適応するとTNF-αが浮かび上がってきた。最近、TNF-αが細胞のがん化を促進することを証明した。従来の化学物質による発がんプロモーターとTNF-αとの関連を明らかにすることを目的として研究を進め、以下の研究成果を得た。
1.化学発がんプロモーターはいろいろな細胞に対し、TNF-αmRNAの発現を誘導し、TNF-αの遊離を促進することを見出した。例えば、オカダ酸はBALB/3T3細胞に、ミクロチスチンやノジュラリンはラット初代培養肝細胞に於いて、TNF-αmRNAの発現を誘導した。
2.オカダ酸の塗布はマウス皮膚に、ミクロチスチンおよびノジュラリンのi.p.投与はラット肝に、それぞれTNF-αmRNAの発現を誘導した。これらの結果は発がんプロモーターの臓器特異性を説明する。
3.ヒト肺がんのプロモーターと推測されるcord factorはプロテインキナーゼCを活性化した。更に、そのi.p.投与は肺組織に於いてTNF-αの産生を促進した。次にcord factorを用いた肺でのプロモーション活性の証明が残されているが、以上の結果より化学発がんプロモーターは、ターゲット臓器の細胞において、TNF-αの生成を促進し、TNF-αを遊離する。このTNF-αが内因性発がんプロモーターとして作用すると理解される。
4.ヒトTNF-αとマウスTNF-αは共に、v-H-rasをトランスフェクトしたBhas42とBALB/3T3の同時細胞培養に於いて、形質転換巣の形成を促進した。しかしBALB/3T3細胞のみに対しては無効であった。
5.SarcophytolAとEGCGはオカダ酸によるBALB/3T3細胞でのTNF-αmRNAの発現とTNF-αの遊離を抑制した。
