研究概要	本研究班は本年度も、ヒト多段階における種々の遺伝子変異の役割の解析に応用可能な、培養細胞を用いての実験系の開発を行い、以下の成果及び知見を得た。 (1)増井はヒト子宮頸部上皮由来のout-growth培養系を用いることにより、偏平上皮細胞の増殖停止機構に関与する遺伝子eti-1を単離することに成功した。 (2)安本はHPV16によるヒトケラチノサイト不死化の系を解析し、細胞同期に関わるp16遺伝子の不死化への関与の可能性を示した。 (3)野田は、子宮頸部多段階発癌過程より多数の培養細胞系の解析から、上皮内癌の基底膜への浸潤におけるメタロプロテアーゼの関与を示した。さらにヒト由来培養細胞における遺伝子操作を容易にする、P1ファージの組換え酵素Creの遺伝子を組込んだ。アデノウイルスの作成に成功した。 (4)難波と清水は、各々ヒト細胞の試験官内発癌系及び染色体移入によるヒト癌細胞の正常細胞化の系を用いることにより細胞癌化に伴うテロメア短縮化消失現象の重要性を示した。 (5)許は、ヒトケラチノサイトの3次元培養系をさらに改良し、創傷治癒過程を試験官内で再現することに成功した。
